The research is focused on understanding the mechanisms of longevity, genome stability and on the studies of exceptionally long-lived mammals.

We can extend human longevity up to 140s and decrease the age-dependent mortality.

Project - Biological immortality

Biological immortality is a state in which the rate of mortality from senescence is stable or decreasing, thus decoupling it from chronological age.
Various unicellular and multicellular species, including some vertebrates, achieve this state either throughout their existence or after living long enough.
A biologically immortal living being can still die from means other than senescence, such as through injury and disease.

Nexedes research is focused on understanding the mechanisms of longevity and genome stability and on the studies of exceptionally long-lived mammals.

Increased DNA repair facilitates greater longevity.

The longer-lived species, humans and heterocephalus expressed DNA repair genes, including core genes in several DNA repair pathways.
These findings suggest that increased DNA repair facilitates greater longevity, and also are consistent with the DNA damage theory of aging and DNA repair transcriptomes.

Aging-induced damage, protein stability and DNA repair pathways.

The longevity has also been attributed to “protein stability”.

Because of their extraordinary longevity, an international effort was put into place to sequence the genome of the heterocephalus.

The transcriptome sequencing revealed that genes related to mitochondria and oxidation reduction are expressed, which contribute to their longevity.

Heterocephalus are living longer than any other rodent, with lifespans in excess of > 40s years.

The mortality rate of the species does not increase with age.

Heterocephalus are highly resistant to cancer and maintain healthy vascular function.

The Gompertz law of mortality
The law of mortality states that the human death rate is the sum of an age-dependent component which increases exponentially with age and an age-independent component.
In a protected environment where external causes of death are rare, the age-independent mortality component is often negligible.
The Gompertz law of mortality describes the age dynamics of human mortality rather accurately in the age window from about 50 to 80 years of age.
At more advanced ages, some studies have found that death rates increase more slowly – a phenomenon known as the late-life mortality deceleration.
The study predicts future longevity records will be broken, reaching into the 140s.

Aging Research Medicine Product – Neuregulin

The Neuregulin (Neregulin) NRG cytokine family comprises four genes that encode a large number of secreted or membrane-bound isoforms. All family members share an EGF-like domain that interacts with the ErbB family of tyrosine kinase receptors.
The NRG gene is the most pleiotropic, encoding more than 14 soluble or transmembrane proteins.
NRG isoforms can be classified into type I, type II and type III isoforms.
The NRG proteins show distinct spatial and temporal expression patterns and play important roles during development of both the nervous system and the heart. Lymphoblastoid cell lines established from blood samples of humans who lived past 100 years (centenarians) have significantly higher activity of the DNA repair protein Poly (ADP-ribose) polymerase (PARP) than cell lines from younger individuals.

The lymphocytic cells of centenarians have characteristics typical of cells from young people, both in their capability of priming the mechanism of repair after H2O2 sublethal oxidative DNA damage and in their PARP capacity.

The partners found that animal species differ dramatically in their aging rates and susceptibility to age-related diseases. Comparative studies on animals with different maximum lifespans can be used to identify the molecular mechanisms that explain this disparity.

The partners found that sirtuin (sirt6) extends mammalian lifespan and promotes healthspan. The sirtuin is maintaining genomic stability through improved efficiency of dna repair, maintenance of heterochromatin and suppression of transposable elements.

The partners are working on genetic and pharmacological approaches that will alleviate age-related pathologies and potential strategy for rejuvenation.
